In vitro study | Nicaraven dose-dependently mildly inhibited the activation of (ADP-ribose) synthetase, possibly directly inhibiting the catalytic activity of poly (ADP-ribose) synthetase in peroxynitrite-stimulated RAW mouse macrophages. In RAW mouse macrophages, Nicaraven partially prevented the peroxynitrite-induced inhibition of mitochondrial respiration and caused a dose-dependent inhibition of nitrite production in bacterial lipopolysaccharide-stimulated macrophage RAW. Nicaraven(0.35 mM) significantly inhibited the maximal aggregation rate induced by adenosine diphosphate (ADP) in healthy volunteer platelets. Nicaraven(1.75 mM) significantly reduced the maximum rate of platelet aggregation induced by collagen. Nicaraven induced dose-dependent inhibition of platelet aggregation in platelets from healthy volunteers and patients with cerebral thrombosis. |
In vivo study | In beagle dogs, Nicaraven inhibits hepatic lipid peroxidation, improves liver function and Systemic Hemodynamics and energy metabolism, and inhibits the release of hepatic enzymes in the blood of hepatic veins, endothelin-1 elevation, histological damage, and neutrophil infiltration into the liver. In male SD rats with transient focal cerebral ischemia, the infarct volume was reduced by Nicaraven(20 mg/kg). In male SD rats with transient focal cerebral ischemia, Nicaraven(60 mg/kg) significantly reduced infarct volume (18.6 and 20.9%). |
