Overview | malachite green, also known as malachite green, is a synthetic dye, CAS No. 569-64-2, molecular weight of 364.92. Glycerol-malachite green staining solution is mainly composed of glycerol, malachite green, deionized water, can be used to check the feces of various helminth eggs. Glycerol-malachite green staining solution is only used in the field of scientific research, and is not suitable for clinical diagnosis or other purposes. |
Procedure | 1. Prepare glass paper strips: Soak the cellophane in glycerol-malachite green staining solution. 2. Place a small amount of stool sample on a newspaper or small paper, and apply pressure on the stool sample with a filter. 3. Collect the screened stool samples by scraping the filter with a blade. 4. Take a plate with a hole and place it in the central position of the slide. Fill the hole with a stool sample when scraping. 5. Remove the plate carefully and leave the stool sample as a short cylinder on the slide. 6. The glass strip was soaked in glycerol-malachite green staining solution, removed and covered with feces. 7. Flip the slide and squeeze the stool sample toward the glass slip on another slide or on an object with a smooth, hard surface, the specimen was spread evenly between the slide and the glass strip. After clarification, the writing of this specification should be able to be read through the smear. 8. Slide the upper slide gently from the side and remove the upper slide to avoid separating from or lifting the glass slide. Place the slide on the test bench with the glass slide facing up. The purpose slides (except for the hook eggs) should be kept at room temperature for one to several hours to make the sample clear. |
note | 1. Smear thickness was appropriate, staining could be performed after fixation. 2, add glycerol-malachite green staining solution to cover the smear is appropriate, not too little, so as not to evaporate and dye precipitation. 3. The dyeing time of hematoxylin, Orange and eosin can be extended appropriately when dyeing in winter, such as when the ambient temperature is low. 4. The fixing liquid should be replaced regularly to avoid insufficient fixation and poor coloring. 5, glycerol-malachite green staining solution should be replaced regularly, so as not to affect the dyeing effect. |
