In vitro study | Embelin is a small molecule inhibitor from the Japanese Purple Golden grass, which binds to the Smac protein and caspsase-9 to the XIAP BIR3 domain. Embelin inhibited the growth of PC-3 and LNCaP cells in a dose-dependent manner with IC50 of 3.7 and 5.7 μm, respectively. At the same time, the toxicity of Embelin to normal Proc and in WI-38 cells was low, and the IC50 was 20.1 μm and 19.3 μm. PC-3 cells were treated with Embelin(25 μm and 50 μm) for 48 hours, 30% and 75% of the cells were apoptotic, 3-fold and 9-fold compared to untreated cells. Embelin is also effective by selectively inhibiting 5-lipoxygenase and microsomal prostaglandin E synthase-1 (mPGES-1 of) inhibition of arachidonic acid biosynthesis (5-LO) with IC50 of 0.06 mM and 0.2 mM, respectively. |
