In vitro study | Brimonidine reduced the progressive loss of ganglion cells by 26% and 15% at doses of 0.5 mg/kg and 1 mg/kg, respectively. Brimonidine prevents further loss of ganglion cells after 10 days of increased intraocular pressure. In the hypertensive retina of the attenuated eye, Brimonidine increases the immune response to GFAP. Brimonidine, but not timolol, was used when intraocular pressure (IOP) was elevated to significantly protect retinal ganglion cells and prevent further cell loss. Brimonidine injection into the eyes of SD rats increased the number of BDNF-positive retinal ganglion cells from 55 to 166 percent. 0.5% Brimonidine,1 drop before, after, or before and after 360-degree argon laser trabeculoplasty significantly reduced the incidence of intraocular pressure. |
In vivo study | In adult rats, Brimonidine(1 mg/kg) significantly protected retinal ganglion cell death induced by ocular hypertension. Brimonidine(0.0001%) caused a loss of about 37% of retinal ganglion cells (RGC) and had no significant neuroprotective effect in adult Sprague-Dawley rats. Brimonidine(0.001 or 0.01) caused retinal ganglion cell (RGC) survival of 76 or 90%, respectively, in ischemic adult SD rats, 0.1%Brimonidine completely prevented retinal ganglion cell (RGC) death. |
