preparation method | The Grignard reagent 5-fluoro-2-methylphenylmagnesium bromide was generated in a single CSTR: A slurry of magnesium (227g,9.4mol) was activated in MeTHF(1L) to prepare mint1 Grignard reagent (1207G), which occurred immediately after the stirring was activated. To compound 1(23%(w/w)) MeTHF was added to the CSTR at a rate of 34.5mL/min, the steady state τ was 0.5 hours, and the vessel temperature was 40 °c. The reaction solution exits through the dip tube and is used as solid magnesium in a Isolation CSTR, then the solution is collected in A accumulation vessel (34.5mL/min) at 1-fold rate. This operation was run as a start/stop operation for more than 5 days with a total processing time of 32 hours. Rest overnight. The yield of 5-fluoro-2-methylphenylmagnesium bromide was 1.68 (69.6 mol). Overall, there was a total of 30 liters of 5-fluoro-2-methylphenylmagnesium bromide Grignard reagent (conversion 97%, yield 95%). |
Application | 5-fluoro-2-methylphenylmagnesium bromide can be used as a pharmaceutical synthesis intermediate. |
