synthesis method | the preparation steps of 6-bromo-2-chloroquinoline are as follows: step 1: 6-bromoquinoline -2(1H)-one (34):33(24.98g,82.7 mmol) and AlCl3(33.10g,248.2 mmol) are ground together in a mortar and pestle to form an intimate mixture. The reaction mixture was transferred to a round bottom flask and quickly heated to melt with a heating gun, and then kept at 110°C for 1.5 hours. The mixture was cooled to room temperature and quenched with ice water. The resulting precipitate was filtered, washed with water and dried to give the title compound, a pink powder, melting point of 272-274°C ([125]272-274 °F)(19.03g,100%), no further purification is required during use. step 2: 6-bromo-2-chloroquinoline (31): reflux heating a mixture of 34(8.52g,38.0 mmol) and phosphorus oxychloride (40 mL,380.0 mmol) for 1 hr. The solution was cooled to room temperature and the excess reagent was removed under reduced pressure. The remaining mixture is quenched with ice water, and the resulting solids are filtered and washed with water. The solid recrystallizes from hexane to obtain the target compound, which is pale pink powder with a melting point of 157-158 ℃([67]157 ℃),(5.34g,58%).
