introduction | 5-fluoroindigo anhydride is a pharmaceutical intermediate, which can be prepared from 2-amino-5-fluorobenzoic acid and triphosgene in one step. 5-fluoroindigo anhydride can be used to prepare a class of 2-(amino) methyl quinazoline -4(3H)-ketone compounds, which can be used as DPP-4 inhibitors for the preparation of antidiabetic and other disease drugs. |
preparation | 2-amino -5-fluorobenzoic acid (13.30g,85.73mmol) is dissolved in 100mL THF, precooled for 0.5h, stirred in an ice bath for 2h, then quickly added bis (trichloromethyl) carbonate (12.72g,42.86mmol), sealed, and continued stirring in an ice bath for 1.5h, transfer to 100 ℃ oil bath for 6h. Concentrate the reaction solution with a rotary evaporator, add 150mL of ether, and filter. The filter cake was washed 4 times with THF-ether (1:10) and dried in vacuum to obtain 14.85g of white powder 5-fluoroindigo anhydride with 95.62% yield. |
