preparation | (1) preparation of 6-chloro-9H-purine: hypoxanthine (25.0g,183.7mmol) is dissolved in 15mL of dimethylaniline in ice bath, then 160mL of phosphorus oxychloride is added dropwise, reflux is completed for 20 minutes, distillation is reduced pressure, the residue is poured into ice water, and ethyl acetate is extracted twice, the pH of the aqueous phase was adjusted to 9 with ammonia water, and then extracted twice with ethyl acetate. The aqueous phase was concentrated under reduced pressure and recrystallized to obtain 26.0g of light yellow solid with 91.6% yield. (2) preparation of 6-hydrazin-9H-purine: heating 6-chlorine-9H-purine (2.0g,12.9mmol), 20mL of ethanol and 1.2mL of 85% hydrazine hydrate to 90°C, reacting for 5 hours, precipitating yellow solid, cooling, suction filtration, washing three times with ethanol and ice water respectively, drying to obtain 1.71g of product, yield: 88.4%. |
