| In vitro study | In Vitro, Everolimus has immunosuppressive activity compared to Rapamycin. Everolimus and immobilized FK 506 competitively bind to biotinylated FKBP12, IC50 is 1.6 nM-2.4 nM, and it acts on the spleen cells of BALB/c and CBA mice to inhibit bidirectional MLR, the IC50 is 0.12 nM-1.8 nM. Everolimus acts on VEGF-induced HUVEC proliferation and bFGF-induced HUVEC proliferation with anti-angiogenic/vascular effects with IC50 of 0.12 nM and 0.8 nM, respectively. Recent studies have shown that Everolimus inhibits all cells and stem cells of the BT474 cell line and primary breast cancer cells, with an IC50 of 156 nM when acting on all cells of primary breast cancer and when acting on all cells of BT474 cells, IC50 is 71 nM. In addition, the combination of Everolimus and Trastuzumab significantly promoted the inhibition of tumor stem cell growth, and the inhibition rate increased by more than 50%. |
