ISOMETAMEDIUM CHLORIDE - Simultaneous determination of triazamidine and phenanthroline chloride residues in bovine tissues by solid phase extraction-high performance liquid chromatography
Abstract:
A solid phase extraction-high performance liquid chromatography (spe-HPLC) method was developed for the simultaneous determination of residues of triazamidine (DIM) and phenanthroline chloride (ISM) in bovine tissues. For different tissue samples, different ratios of water-acetonitrile solution were used to extract the drug, and the Oasis WCX solid phase extraction column was combined to purify and enrich. The drug was separated on a Spherisorb CN column (250 × 4.6, 5 μm) with the mobile phase of acetonitrile and 0.05 mol/L ammonium formate solution (pH 2.4) by gradient elution, the UV detection wavelength was 380 nm. The limit of detection (S/n≥ 3) of DIM and ISM in bovine tissues was 0.01 mg/kg, and the limit of quantification (S/n≥ 10) was 0.025 mg/kg. DIM and ISM in bovine tissues had good linear relationship in their respective linear ranges (r≥ 0.999 3). The recoveries of DIM and ISM at low, medium and high spiked levels ranged from 82.2% to 97.6%, and the intra-day and inter-day precision ranged from 0.3% to 5.2%, respectively (n = 5). And 1.3% ~ 5.2%(n = 15). The results showed that the established method had good repeatability, high sensitivity and simple operation, and was suitable for the detection of DIM and ISM residues in bovine tissues.expand
