Abstract:
objective to study the expression changes of metastasis-associated genes in human highly metastatic lung cancer cells before and after drug treatment, and to further reveal Icariin (ICA). And the mechanism of the anti-metastasis effect of PJA of Jinan Pseudomonas preparation. Methods flow cytometry and reverse transcription-PCR(RT-PCR) were used to detect the protein and m RNA levels of metastasis-associated genes c-m yc,Tiam -1, nm23-h1. Results after treated with PJA and ICA for 48 h, the m RNA of c-m yc,Tiam -1 gene in PG cells were decreased in different degrees, while the level of nm23-h1 m RNA was increased in different degrees, after 96 h of drug treatment, the change of gene level was reflected to the expression product-protein level. The ascending and descending trends of c-m yc and nm23-h1 gene proteins were consistent with their m RNA levels (P 0.005), and the changes of the two levels were the most significant in the synergistic group. Conclusion PJA,ICA can inhibit the expression of metastasis-inducing gene c-m yc,Tiam -1 and promote the expression of metastasis-inhibiting gene nm 23, which play an anti-metastasis role.

Abstract:
icariin, a traditional Chinese medicine monomer, is a new type of biological response modifier and differentiation inducing agent. In order to further explore the anti-cancer mechanism of icariin, we use MTT, radioimmunoassay, flow cytometry, adhesion test, movement invasion experiments and other means were tested.
