in the early 70s, Shida et al. extracted three water-soluble polysaccharides from Lentinus edodes by trichloroacetic acid extraction, methanol precipitation and other methods, one of which was composed of Gal,L-Fuc and Man (molar ratio 6:1.5:1). this is another anti-tumor substance reported for the first time in Lentinus edodes except β-glucan. Sasaki also reported that besides the macromolecule lentinan with a relative molecular mass of 1 million, another small molecule lentinan with a relative molecular mass of 16000 also has anti-tumor activity.

Other polysaccharides in Lentinus edodes are composed of glucose, galactose, mannose, L-arabinose and xylose, and there are also α-mannosides derived from the mycelium of submerged fermented Lentinus edodes.
In recent years, people have carried out research on chemical modification of lentinan to find more effective polysaccharide substances. For example, 10 μg/mL sulfated lentinan can effectively protect HIV-infected MT-4 cells, and 100 μg/mL can inhibit RT activity and the generation of synaptic cells. Its inhibitory effect on stress transcriptase is higher than that of sulfated dextran. Through 13C-NMR and other measurements, it is proved that the introduction of sulfuric acid base leads to the physical and chemical properties of lentinan and the conformational changes of the main body, resulting in the different immune activity of the derivatives.

The Sevag method was used to unfold with chloroform and 1-butanol, and 3 times the volume of ethanol was added to precipitate it. Collect the precipitate, wash twice with methanol, wash once with ether, and dry in vacuum at room temperature and in the presence of calcium chloride to obtain lentinan. 31g of lentinan can be extracted from 200kg of raw materials.
