pharmaceutical excipients
pregelatinized starch, also known as compressible starch, is a new type of pharmaceutical excipients, which have been collected in British, American, Japanese and Chinese pharmacopoeia. China successfully developed it in 1988 and has now been supplied to the market in large quantities. Domestic compressible starch is a partially pregelatinized product (fully pregelatinized starch is also called α-starch), which is equivalent to Starch RX1500 of foreign Colorcon companies. This product is a multifunctional auxiliary material, which can be used as a filler. It has good fluidity, compressibility, self-lubricity and dry adhesion, and has a good disintegration effect. If used for direct compression of powder, the amount of magnesium stearate should not exceed 0.5% to avoid softening effect.
source
pregelatinized starch is a derivative of starch. First of all, let's learn about starch. Starch has a wide range of sources, mainly from mature seeds of wheat, corn, rice and tubers of potato and cassava. As early as more than 100 years ago, starch was widely used in the production of solid preparations as a pharmaceutical excipient. Due to the poor compressibility of starch, it needs to be combined with appropriate amount of sugar powder or dextrin when applied. Researchers have carried out modification research on starch, resulting in the emergence of a new product of pregelatinized starch.
method
pregelatinized starch is mainly to add water to corn starch to make it into a slurry, and then use heating or mechanical method to destroy the long chain of glucose molecules in starch, so that all or part of it is broken and finally dried and crushed. Part of the pregelatinized starch is generally made of wet starch by mechanical pressurization.
classification
pregelatinized starch is divided into fully pregelatinized starch and partially pregelatinized starch (partially pregelatinizedstarch) according to the degree of pregelatinization. The degree of pre-gelation is different, and the function is also different. Most of the domestic company's products are full pre-gelatinized starch, Calecon's product Starch1500 is part of the pre-gelatinized starch trade name Sunda. Fully pregelatinized starch can be completely dissolved in cold water. When used in wet granulation, starch slurry can be directly prepared without heating. Therefore, it can be directly added to the granulation equipment together with the main medicine and excipient, and then water is added as wetting agent. Partially pregelatinized starch (PPS) contains soluble (gelatinized) and insoluble components. Those insoluble ingredients contain complete starch granules, and the soluble ingredient is modified starch, so it has disintegration characteristics in addition to adhesion. In addition, the multi-granular pregelatinized starch with larger particles showed better fluidity than the natural starch.
traditional applications
It is mainly used as a filler and adhesive in the prescription of oral capsules and tablets. Application one as a binder used in the wet granulation process, can be prepared for starch slurry or water wetting use, our company's application of the main varieties of roxithromycin particles, estazolam tablets and so on. Application 2 as a filler used in the wet granulation process, the main varieties of application are Zhou Xiao sulfamethamine tablets, roxithromycin tablets, cefaclor tablets and so on. Application 3 is used as a filler in full powder compression tablets and used in combination with microcrystalline cellulose or lactose. The application varieties include alendronate sodium tablets, levamlodipine maleate tablets, and enalapril maleate tablets Wait. Application four as a filler used in capsule filling, the application of varieties such as norfloxacin capsules.
