in this paper, the essential oil contained in the natural flavor plant sage was extracted by steam distillation, and the essential oil composition was analyzed by gas-gas mass spectrometry (GC/MS). The results showed that 62 components were isolated, 51 of them were identified, and their relative contents were determined. The separation effect of this method is good, the identified components are more accurate, and it has certain reference value for the development and application of sage essential oil.

Salvia (Salviaofficinalis) is a plant of the genus Salvia in the family Lamiaceae. It has a wide range of physiological activities. Modern pharmacological experiments have proved that Salvia has the functions of dilating coronary arteries, increasing coronary flow, improving microcirculation, and protecting the heart. It can inhibit and relieve platelet aggregation, improve the body's hypoxia tolerance, and has anti-tumor and antiviral activities. It is an important medicinal plant. In this paper, the extraction, purification and antioxidant activity of sage essential oil were studied, and it is possible to discover and develop new functions and new uses of sage. (1) With sage as raw material, supercritical CO2 fluid extraction and steam distillation were used to extract its essential oil. With the extraction rate as an index, the optimal extraction conditions of the two extraction methods were determined: supercritical CO2 extraction method: sage particles 40-60 mesh, extraction temperature 50 ℃, extraction pressure 15MPa, carbon dioxide flow rate 20L/h, extraction time 120min, separation temperature 40 ℃. The extraction rate of sage essential oil extracted by supercritical CO2 fluid was 6.01±0.13%. Steam distillation method: sage particles 40-60 mesh, distillation time 1.5h, solid-liquid ratio 1:8, soaking time 1.5h, distillation aid 2%(w/w)NaCl, microwave-assisted extraction time 30s. The extraction rate of sage essential oil extracted by steam distillation was 3.04±0.15%. (2) The physical and chemical properties and composition content of sage essential oil extracted by the two methods were determined, and the physical and chemical properties of sage essential oil extracted by steam distillation were the closest to Spanish sage oil, but the composition was quite different; the composition of sage essential oil extracted by supercritical extraction was close to that of Spanish sage oil, however, its relative density and refractive index are slightly beyond the standard range. (3) The free radical elimination rate of sage essential oil extracted by the two methods was measured to evaluate its antioxidant activity: the results showed that the sage essential oil extracted by the two methods in the three antioxidant systems of DPPH ·,· OH and O2-· had obvious antioxidant activity, the antioxidant activity of supercritical extraction essential oil is slightly higher than that of steam distillation. (4) The supercritical extraction of sage essential oil was purified by silica gel column chromatography. The results showed that the purification effect was best when the eluent ratio was V petroleum ether: V ethyl acetate = 9:1. After GC/MS analysis, the main components of sage essential oil were effectively enriched, and the total content of α-pinene, α-butrone, camphor, linalool, linalool acetate and caryophyllene increased from unpurified 8.5% to 23.3%.
