In vitro study | IPA-3 allosteric inhibitors of p21-activated kinase 1(Pak1) that are non-ATP competitive. IPA-3 inhibition of Cdc42 stimulated Pak1 autophosphorylation at the Thr423 site. IPA-3 also inhibited sphingosine-dependent Pak1 autophosphorylation. IPA-3 does not target the cysteine residues that act on the exposed pak1. A disulfide bond of IPA-3 is critical for inhibition of Pak1 and IPA-3 inhibition of Pak1 is abolished by the reducing agent dithiothreitol (DTT). IPA-3 inhibits Pak1 activation by different activators, but does not inhibit pre-activated pak1. IPA-3 inhibits PDGF-stimulated activation of Pak in mouse embryonic fibroblasts. IPA-3 inhibition of Pak1 activation by covalent binding to the regulatory domain of pak1. IPA-3 is covalently bound to Pak1, and this effect is time and temperature dependent. IPA-3 inhibits the binding of the Pak1 activator Cdc42. IPA-3 binds directly to the autoregulatory domain of pak1. IPA-3 acting on cells, reversible inhibition of PMA induced membrane cracking. |
