use 4-amino -2-methyl-10H-thiophene [2,3-b][1,5] benzodiazepine hydrochloride is yellow to dark beige solid at normal temperature and pressure, which is common in the synthesis of olanzapine tablets, a drug molecule for the treatment of schizophrenia.
synthesis method in a 500 ml round bottom flask equipped with a mechanical stirrer and thermometer, put 10.0g (0.0386 mol) 5-methyl -2-((2-nitrobenzene) amino) thiophene -3-nitrile (1) and 100 ml ethanol. Then 7.4g (0.062 mol) of tin powder and 84 ml of 4M hydrochloric acid were added, and the mixture was stirred at 50 ℃ for 0.5 hours. The temperature of the reaction mixture was raised to 85°C and the stirring was continued for 4 hours. The mixture was then cooled to 25°C and stirred for a further 8 hours, which resulted in the precipitation of solids. The precipitated crystals were collected, washed with 20mL of ethanol, and dried in a vacuum. 9.8g (96%)2 hydrochloride is a light yellow solid. mp. above 300 ° 8(HPLC>98.5%).
