Application

7-bromo-3, 4-dihydroquinazolin-4-one is a quinazolone compound. Quazolidone is a kind of nitrogen-containing heterocyclic compound with good biomedical activity, due to its extensive biological and pharmaceutical activities.

preparation

step 1: synthesis of methyl 2-amino -4-bromobenzoate at room temperature, 2-amino -4-bromobenzoic acid (12.0g,55.5mmol) and MeOH(200mL) are added to a single-mouth bottle, concentrated sulfuric acid (12mL) is added dropwise at 0 ℃, and the reaction is terminated after 48h at 80 ℃, the reaction was cooled to room temperature, concentrated, the residual liquid was poured into ice water slowly, the pH was adjusted to 8 with saturated NaHCO3, extracted with EA, washed with organic phase, washed with saturated NaCl, dried with anhydrous Na2SO4, filtered and concentrated, column chromatography (PE/EA system) to obtain 11g of white solid with 87% yield. Step 2: Synthesis of 7-Bromo -3,4-Dihydroquinazoline -4-one At room temperature, 2-amino -4-bromobenzoate methyl (2.5g,10.86mmol) and formamide (18mL) are added to a 30mL microwave tube, the microwave reaction at 200 ℃ is terminated after 1h, two batches are cooled to room temperature, EA is added for dissolution, water is washed twice, and anhydrous Na2SO4 is dried, filter concentration, crude products are directly used in the next step.
