preparation methyl 3-aldehyde indole-5-formate as a derivative of indole-3-formaldehyde has not been reported in the literature. According to the molecular structure characteristics of compound 1, methyl 3-methyl -4-nitrobenzoate was prepared by esterification of 3-methyl -4-nitrobenzoate with 3-methyl -4-nitrobenzoate. compound 2 was condensed with N,N-dimethylformamide diethyl acetal (DMFDEA) and piperidine after being reduced by iron powder and glacial acetic acid to obtain methyl indole -5-formate. finally compound 4 was synthesized by Vilsmeier-Hacck reaction with phosphorus oxychloride and N,N-dimethylformamide, the process route is simple to operate, high yield, suitable for industrial production.
