| overview | bisphosphonates (Bisphosphonates,BPs) are a new class of drugs used for various bone diseases and calcium metabolic diseases. they can specifically combine with hydroxyapatite in bone to inhibit osteoclast activity, thus inhibiting bone absorption. they are widely used clinically to treat osteoporosis, deforming osteitis, hypercalcemia and tumor-related bone diseases. Bisphosphonate drugs such as zoledronic acid have a powerful anti-bone resorption effect and its mechanism. They directly change the morphology of osteoclasts, inhibit the formation of actin, and interfere with the adsorption and reabsorption of bone by osteoclasts; It also inhibits the release of peptides in the bone matrix, breaks the vicious circle called "seed and soil mechanism" between tumor and bone formation, and inhibits the growth of tumor cells. Zoledronic acid EP impurity D is an impurity produced during the synthesis of zoledronic acid. |

Filtration, saturated salt water washing filtrate three times, anhydrous sodium sulfate drying, distillation of the organic phase under reduced pressure at 25 ℃ until the droplets no longer drip out, to obtain oil, namely compound Ⅲ intermediate 1;(2) Preparation of Zoledronic Acid EP Impurity D Compound Ⅲ Intermediate 1 is 8.4g(0.05mol) dissolved in 100mL of water, concentrated hydrochloric acid adjusts the pH value of the solution to 2.0, reflux at 120 ℃ for 5 hours, after the reaction is over, the obtained reaction liquid is directly distilled under reduced pressure at 25 ℃ until the droplets are no longer dripped out, and 300mL of acetone is added to obtain a white solid, filtration, and recrystallization of isopropyl alcohol to obtain a white crystal, namely zoledronic acid EP impurity D. |
