after aniline reacts with hydroxylamine and chloroacetaldehyde hydrate, isatin is catalyzed by sulfuric acid to obtain isatin with 63.4% yield. Indigo, excess hydroxylamine hydrochloride, sodium acetate trihydrate and water, stirred vigorously at 60 ℃ and left overnight at room temperature. Filtration, ethanol-water recrystallization to obtain indigo -3-oxime with 92% yield. Indiat-3-oxime is heated to melt, and it decomposes vigorously to form o-aminobenzonitrile, which is dissolved and collected with ether. The product is distilled under reduced pressure after distillation of ether with 86.8% yield, boiling point 112~114 ℃/0.8kPa and melting point 48~50 ℃. O-aminobenzonitrile is dissolved in cyclohexanone, equimolar anhydrous zinc dichloride is added, and the reflux is stirred. Cool and filter, the solid is washed with ether several times, and then suspended in water. Add 30% sodium hydroxide to strong alkaline and extract with dichloromethane. The extract was washed, dried, concentrated and recrystallized with toluene to obtain yellowish flake tacrine crystals with 92.6% yield and melting point 182~184 ℃.
