| Introduction | 3-chloro-5-fluorobenzonitrile is a nitrile organic compound, which can be used as a pharmaceutical synthesis intermediate, it can be obtained from 1-bromo-3-chloro-5-fluorobenzene by two-step preparation. 3-chloro-5-fluorobenzonitrile can be further prepared by strong base hydrolysis to 3-chloro-5-fluorobenzoic acid. |
| prepare | in 70 ml N, 25.0G of 1-bromo-3-chloro-5-fluorobenzene (120 mmol), 8.45G of zinc cyanide (72 mmol), 235 mg zinc powder (3.6 mmol), 1.5g 1,1 '-bis (diphenylphosphino) ferrocene] palladium (II) dichloride A 1: 1 complex with dichloromethane (1.8 mmol) was heated to reflux for 1 hour. After cooling, the reaction was diluted with ethyl acetate and extracted with water and brine. Chromatography on silica gel gave 15.9g (85%) of 3-chloro-5-fluorobenzonitrile. |
