| In vitro study | 4-Diethylaminobenzaldehyde (DEAB) is reversible competitive inhibitor of ALDH1 (competitive with the aldehyde substrate) with a K i value of 4 nM, and irreversibly inactivates ALDH7A1 via formation of a stable, covalent acyl-enzyme species. The Temozolomide (TMZ)-resistant cell line LN18 is treated with a combination of TMZ and 4-Diethylaminobenzaldehyde for 10 days. As indicated by colony formation assay, the clonogenic capacity of LN18 cells exposed to TMZ in combination with 4-Diethylaminobenzaldehyde (DEAB) is significantly attenuated. |
| Risk Codes | R36/37/38 - Irritating to eyes, respiratory system and skin. R50/53 - Very toxic to aquatic organisms, may cause long-term adverse effects in the aquatic environment. R21/22 - Harmful in contact with skin and if swallowed. |
| Safety Description | S26 - In case of contact with eyes, rinse immediately with plenty of water and seek medical advice. S36 - Wear suitable protective clothing. S61 - Avoid release to the environment. Refer to special instructions / safety data sheets. S60 - This material and its container must be disposed of as hazardous waste. S36/37 - Wear suitable protective clothing and gloves. |

| Biological activity | 4-Diethylaminobenzaldehyde is a reversible aldehyde dehydrogenase (ALDHs) inhibitor, and the Ki for ALDH1 is 4 nM. 4-Diethylaminobenzaldehyde had a strong antiandrogen effect (IC50=1.71 μM). |
| Use | Used as a dye intermediate for the synthesis of cationic dyes Flavor synthesis. Analytical reagents, organic intermediates. |
| production method | diethylaniline, urotropine and 90% ethanol are mixed evenly, heated and refluxed, glacial acetic acid and 90% formic acid are added, and kept warm overnight. The reaction liquid is poured into the pre-cooled hydrochloric acid, filtered, and the crude product is recrystallized by acid-base and ethanol to obtain p-diethylaminobenzaldehyde. |
