| preparation | methionine oxidation: methionine is dissolved in 5M perchloric acid aqueous solution, 0.05M ammonium molybdate, hydrogen peroxide, water and pyridine are added for neutralization (PH = 7), acetone is added for precipitation, purification and drying to obtain methionine sulfone (II). Among them, methionine: 5M perchloric acid: water: 0.05M ammonium molybdate: 30% hydrogen peroxide: water: acetone = 0.0336mol: 7ml: 45ml: 3ml: 7g: 70ml: 240ml. |
| detection | HPLC method for simultaneous direct determination of methionine sulfoxide (MSO) and methionine sulfone (MSO2) impurities in compound amino acid injection (18AA). TitankC18 chromatographic column (4.6mm × 250mm,5 μm), 10mmol/L(NH4)2HPO4 buffer (containing 10mmol/L1-sodium octane sulfonate, pH adjusted to 1.9 with phosphoric acid) as mobile phase A, acetonitrile as mobile phase B, gradient elution, flow rate 1.0 mL/min, detection wavelength 205nm, injection volume 100 μL, column temperature 4 ℃. The results show that methionine sulfoxide and methionine sulfone can be effectively separated under the optimized conditions. The detection limits (S/N = 3) of methionine sulfoxide and methionine sulfone were 0.09 μg/mL and 1.35 μg/mL, respectively; the quantitative limits (S/N = 10) were 0.31 μg/mL and 4.51 μg/mL, respectively; the linear relationship between methionine sulfoxide and methionine sulfone was good in the range of 0.31~201.76 μg/mL and 4.51~225.72 μg/mL, respectively; the recoveries were 99.1% ~ 102.2% (methionine sulfoxide) and 98.2% ~ 101.2% (methionine sulfone), respectively. This method can be used to detect methionine sulfoxide and methionine sulfone in compound amino acid injection (18AA) in conventional laboratory and pharmaceutical industry. |
