| Physical and Chemical Properties | Chemical properties light red or light yellow freeze-dried powder, soluble in water, the optimum pH value is 7.0-7.5 (pig kidney, amylase) or 7-8 (mirotrase). Stability: pure enzyme can be preserved for a long time after freeze-drying. After toluene is added to the crude enzyme solution, it can be stored in the refrigerator for 1-2 days in a short period of time. Crude enzyme preparation can exist stably at 5 ℃. When pH is 7, the enzyme aqueous solution keeps its activity unchanged at 70 ℃ for 60min, and when pH is less than 5, it is rapidly inactivated at 70 ℃. Requirements for the chemical structure of the substrate: monocarboxyl amino acid; the amino group on the α-carbon atom in the amino acid is acylated, and the hydrolysis speed of the acylation group is different; the chemical structure of the substrate is different from the enzyme of different sources. Enzymatic reaction: N-acylated-L amino acid H2O; fatty acid anion L-amino acid. |

Acylase 1 is an extremely stable enzyme in the dry state. In solution, the enzyme is stable at high temperatures (70° C) at pH 7, but below pH 5 it is rapidly and irreversibly inactivated. The enzyme shows a high degree of optical specificity toward its substrates. For this reason, it has been used extensively for the resolution of racemic amino acids.

9012-37-7 - Production method
pig kidney, amylase or Aspergillus oryzae are used as raw materials, after homogenization and centrifugation to deprecipitate, ammonium sulfate is used for graded precipitation, water dialysis and centrifugation to obtain crude enzyme solution, and then acetone and ethanol are used for graded precipitation and DEAE-cellulose column chromatography purification to obtain the product.
