2-amino-5-nitrotoluene was dissolved in 2.5L of glacial acetic acid and cooled, A solution of 25g of sodium nitrite and 60ml of water is added and the reaction temperature does not exceed 25 °c. The resulting yellow precipitate was filtered off, the reaction solution was placed for 3 days and then concentrated under reduced pressure. The residue was pasty with 200ml of water, filtered, the filter cake was washed with cold water, dried, and recrystallized with methanol, 42-47g
5-nitroindazole was obtained in a yield of 72%-80%. N,N-dimethylformamide solution and 5-nitroindazole were added to a three-port reaction flask, and the mixture was stirred for 30 minutes. Methyl iodide was slowly added dropwise at 0 ° C., and the mixture was heated to room temperature for reaction. The reaction progress was detected by thin layer chromatography. After the reaction was complete, ethyl acetate was added and washed with water for 3-4 times, concentration gave the product 5-nitro-3-methyl-1h-indazole. 5-nitro -3-methyl -1H-indazole and methanol were added into a 2L single-necked bottle, and Pd/C(10g,Pd: C = 10: 1) was added after stirring and dissolving, after removal of air, hydrogen gas was introduced. After stirring at room temperature for 4 hours, the reaction was complete. The Pd/C was removed by Suction filtration, and the filtrate was distilled off to obtain a crude solid, which was dissolved in 500ml of ethyl acetate and washed with 200ml of water. The organic phase was dried over anhydrous sodium sulfate, and the filtrate was distilled off the solvent, the residue was slurried with n-hexane and dried to give the target compound 3-methyl-1h-indazol-5-amine.
