| In vitro study | In the hippocampus of chronically stressed-electrocuted mice, Agomelatine completely normalized stress-affected cell survival and partially reversed decreased doublecortin expression. |
| In vivo study | In transgenic mice, Agomelatine effectively reversed the forced swim test, as well as behavioral changes in the elevated plus maze. Agomelatine also significantly accelerated the adjustment of the diurnal cycle of temperature and activity, followed by an induced content phase shift. In the ventral hippocampus (VH) of adult rats, an area associated with mood disorders, Agomelatine enhances cell proliferation and neurogenesis. Agomelatine increases the proportion of mature and immature neurons and enhances neurite outgrowth of granule cells in adult rats, suggesting accelerated maturation. Agomelatine also activates multiple cellular signals (extracellular signal-regulated kinase 1/2, protein kinase B, and glycogen synthase kinase 3β), these signals are antidepressant regulation and are involved in the control of proliferation/survival. In unfamiliar pairs of rats exposed to a novel environment, Agomelatine increased the time spent participating in social activities. In the ventral dentate gyrus of the Rat, Agomelatine increases cell proliferation and neurogenesis, an area involved in affective responses that is consistent with the antidepressant anxiolytic properties of Agomelatine. In the entire dentate gyrus of the Rat, Agomelatine increased the survival of newborn neurons. |
