CTP - Production method
Fermentation with CTP as raw material
Fermentation and protein removal take 6.5kg of potassium dihydrogen phosphate, add appropriate amount of water to dissolve, add 0.6kg of magnesium chloride and 17.5kg of glucose, stir to dissolve it, take 2.5kg of CMP2.5kg and add water to dissolve it, adjust pH = 7 with sodium hydroxide, add water to 70L, heat to 50-60 ℃ under stirring, add 180L of yeast solution, keep 30-32 ℃ for 3h, add hydrochloric acid to adjust pH =. Add trichloroacetic acid, adjust pH = 2.5, centrifuge to obtain clear liquid.

Salt removal, precipitation and purification Add the clear liquid to strongly acidic styrene cation exchange resin (001 × 7), stir and adsorb for 15min, make the pH be 1.5-1.7, let stand for 30min, filter, add ethanol to the filtrate to the concentration of alcohol be 75%, let stand overnight, filter, add water to dissolve the filter cake to make the concentration be 25%, add diatomite 1/5 the volume of the filter cake, stir and filter, and collect.
Supernatant [001 × 7 resin] & rarr;[pH1.5-1.7, 15min] filtrate [ethanol] & rarr;[8h] sediment [water, diatomaceous earth] & rarr;[stirring, filtering] filtrate
Adsorption, elution and decolorization: dilute the filtrate with water to a solution of 0.5%-0.6%, adjust Ph = 2-2.5, adsorb with strong alkaline quaternary ammonium type I anion exchange resin (717), wash with water for 8 hours, and then elute ADP with 0.03mol/L sodium chloride solution of pH 2. When the absorbance of the solution at the wavelength of 280nm is greater than 0.4, wash with water at pH2 for 4 hours, finally elute ATP with 1mol/L sodium chloride solution at pH2, add 1‰ amount of activated carbon to the eluent, stir, decolorize, filter and collect filtrate.
Filtrate [717 resin] & rarr;[pH2-2.5] adsorbate [sodium chloride] & rarr;[pH2] elution [activated carbon] & rarr;[40min] filtrate
Precipitate and dry filtrate adjusted Ph = 3-5, add 3 times the amount of ethanol, stand, filter, wash the filter cake with anhydrous ethanol and anhydrous ether, vacuum drying, that is, cytidine disodium triphosphate.
Filtrate [ethanol] & rarr;[pH3.5] precipitate [anhydrous ethanol, anhydrous ether] & rarr; Cytidine disodium triphosphate.
