| introduction | N-fluorene methoxycarbonyl-L-alanine is also called (S)-2-((((9H-fluoren-9-yl) methoxy) carbonyl) amino) propionic acid, which is an amino acid derivative, it can be obtained by L-alanine protected by Fmoc. There are reports that it can be used to prepare an aromatic heterocyclic derivative (S)-5-(1-aminoethyl)-2-(3-(cyclopropyl methoxy)-4-methoxyphenyl)-N- (2, 4-difluorobenzyl) thiazol-4-formamide hydrochloride, the pharmaceutical composition containing the compound has a good affinity for PDE4, especially for chronic respiratory obstruction, it has a good therapeutic effect. |
| preparation | Na2CO3(29.8g,280.9mmol) and 9-fluorene methyl-N-succinimide carbonate (28.4g,84.27mmol) were added to L-alanine (5g,56.18mmol) of water/dioxane (v/v = 1/1,400mL) solution, stirring at room temperature for 18h, filtering, Et2O extraction (100mL × 3), water phase acidification with concentrated hydrochloric acid, ethyl acetate extraction (150mL × 3), combining organic phase, drying with Na2SO4, concentration, 17.3g white solid, yield: 99%. |
