Nattokinase - Cloning and expression of Nattokinase gene

Abstract:
The Nattokinase gene was amplified from Bacillus subtilis genomic DNA secreting Nattokinase by PCR, and its nucleotide sequence was determined. The expression vector of Nattokinase gene was constructed by Gene Recombination technology and expressed in E. Coli. SDS-polyacrylamide gel electrophoresis showed that the expressed protein accounted for 15.2% of the thalli protein, and the expression product had thrombolytic activity by agarose-fibrin plate method.

Nattokinase - Optimization of liquid fermentation conditions of Nattokinase

Abstract:
The liquid fermentation production of Bacillus subtilis (BSN) was studied. The effects of fermentation conditions (temperature, inoculum size, initial pH, liquid volume) and medium composition (carbon source, nitrogen source, metal ions, etc.) on the yield of Nattokinase were investigated; on the basis of optimizing the fermentation conditions and culture medium, the amplification experiment of 5L fermentor was carried out, and the yield of enzyme was 2 25 IU/mL.

Nattokinase - Introduction
The activity test method 1. the preparation of the sample solution and dilutes the sample to an appropriate concentration with a diluent, which is the liquid to be tested. 2. test method operation 1. mix 1.4 mL Borate buffer (0.05 mol/L;pH 8.5) and 0.4 mL Fibrinogen solution (0.72%) evenly and place them in a water bath at 37 ℃ for 5 minutes. 2. add 0.1 mL Thrombin solution and mix well. 3. After 10 minutes, add 0.1 mL of the solution to be tested immediately, mix it and place it in a 37 ℃ water bath. 4. the reaction shall be carried out under the condition of mixing the solution evenly. after 60 minutes, 2 mL TCA solution (0.2 mol/L) shall be added immediately to terminate the reaction at 37 ℃ for 20 minutes. 5. Place the mixture in a microcentrifuge tube for 15000 rpm for 5 minutes. 6. Carefully take 1 mL of supernatant and measure the absorbance value (A τ) of OD275. 3. calculation method 1 unit activity (1 FU): In this test method, the OD275 absorbance value is increased by 0.01 enzyme amount per minute. Nattokinase enzyme activity (FU/g)=(Aτ-AB)/ 0.01 × 1/60 × 1/0.1 × DA τ: absorbance AB of the liquid to be tested: absorbance d of the blank group: dilution multiple of the sample

Nattokinase - Study on the anticoagulant effect of Nattokinase

Abstract:
objective to study the activity and anticoagulant effect of Nattokinase (NK). Methods the activity of NK was determined by fibrin plate method. The whole blood clotting time, plasma re-Calcium Time, thrombin time, rat thromboplastin time, thrombin-induced fibrin coagulation time; The prothrombin time of rats was determined by Quick method. Results the activity of solid powder NK was 5 6 5 5 10U/g, which was 3.13 times of that of urokinase, and the activity of liquid NK was 15 2 34U/ml;NK can significantly prolong the rabbit whole blood coagulation time, plasma re Calcium Time, thrombin time, rat thromboplastin time, thrombin induced fibrin clotting time, rat prothrombin time. Conclusion NK has influence on the three stages of coagulation process.
