Preparation | Add (S)-(4-benzylpiperazine -2-yl) methanol (15K)(10.3g,50mmol) and methanol (100mL) to a 200mL round bottom flask. add carbon-loaded palladium hydroxide (3.0g) and circulate the flask with hydrogen and vacuum five times. The flask was stirred overnight with atmospheric hydrogen at room temperature. The reaction was filtered through diatomite with a Brinell funnel. Cool the filtrate to -15°C. Boc anhydride (10.9g, 1 eq) was slowly added to the methanol solution. When the addition is complete, heat the flask to 0°C and stir for 2 hours. Heat the flask to room temperature and remove the solvent under vacuum. The residue was purified by column chromatography using 4:2 ethyl acetate methanol to obtain 15L of oil (11.01g,85%). |
