Preparation | 1) 0.95 g4-(hydroxymethyl)-2, 6-dichloropyridine and 326mg sodium azide were added to 5.00mL of N,N-dimethylformamide, and the mixture was stirred at 70°C. Concentrate the reaction solution, add water to the residue, and extract the mixture with chloroform. The organic layer was dried with sodium sulfate, the sodium sulfate was filtered out, and the organic layer was concentrated to obtain 810mg of azide. 2) Add 1.35g of azide and 1.94g of triphenylphosphine obtained in the above steps to 30.00mL of tetrahydrofuran and 3.00mL of water, and stir the mixture overnight at room temperature. The reaction solution was concentrated and the residue was purified by silica gel rapid chromatography (elution with methanol-chloroform) using a rapid automatic purifier (Biotage AB/Isolera TM) to obtain 220mg of the 2,6-dichloropyridine-4-methylamine compound. |
