In vitro study | In vitro experiments, SB225002 inhibited groα-stimulated calcium mobilization and effectively inhibited IL-8 and groα-induced chemotaxis of human and rabbit neutrophils. SB 225002 substantially reduced the level of phosphorylated ERK1/2 and reduced cell differentiation in WHCO1 cells. SB225002 also exhibits anti-tumor activity as a microtubule inhibitor. In vivo study | In rabbits, SB225002 selectively blocked IL-8-induced neutrophil flanking. SB225002 (1 mg/kg I. p.) inhibits the growth of subcutaneously implanted tumors in a mouse intrahepatic cholangiocarcinoma model. In addition, SB225002 also exhibited a long-acting anti-inflammatory effect and reduced TNBS-induced colitis in a mouse model.
