Introduction | p-fluoroiodobenzene, also known as 4-fluoroiodobenzene, is an organic intermediate. It has been reported in the literature that it can be obtained from 4-fluoroaniline through diazotization reaction or obtained from fluoroiodobenzene. |
use | 4-[18F] fluoroiodobenzene is a widely used building block in 18F radiochemistry, used for various transition metal-mediated C- C and C- N cross-coupling reactions. The cross-coupling reaction of 4-fluoroiodobenzene with potassium pentafluorophenyl trifluoroborate in toluene in the presence of different palladium catalysts and silver oxide was studied. |
preparation method | add 10ml of glacial acetic acid and 5ml of acetic anhydride into a 100ml round bottom flask, add magnetic stirring, cool to 5-10 ℃ in an ice water bath, add 1.7g(13.4mmol) of iodine and 0.48g(2.25mmol) of sodium periodate, keep the temperature unchanged, slowly add 5ml of concentrated sulfuric acid dropwise, after 10min, 1.5g(15.6mmol) of fluorobenzene was slowly added, the reaction temperature was adjusted to room temperature, the reaction mixture was continuously stirred for 5h, 1g of sodium sulfite dissolved in 40ml of ice water was added, stirring was stopped after 10min, extraction was carried out with 2 × 40ml of dichloromethane, the combined organic layer was washed with 3 × 50ml of saturated sodium bicarbonate solution, and the organic layer was dried with anhydrous sodium sulfate. The solvent was distilled under reduced pressure, and the product was yellow oil to obtain 2.46g of the product with a yield of 71%. |
