Preparation | The solution of 3g(42mmol) 3-butene-1-ol in 10mL anhydrous pyridine is cooled in an ice bath. Toluenesulfonyl chloride (7.9g,42mmol) was added. After stirring for 3 hours, pour the mixture into 30mL of ice/concentrated HCl4/1v/v solution, extract with 60mL of ether and dry with MgSO4 in the refrigerator overnight. Filter the mixture, evaporate the ether, and dissolve 1.56g(6.9mmol) of ethyl acetamide dipropionate in 10mL of anhydrous THF at room temperature under N2. Potassium tert-butoxide (0.80g,7mmol) was added under vigorous stirring. The mixture was heated at 60°C for 2 hours. 3-butene-1-ol 4-methylbenzene sulfonate (1.5g,6.9mmol) was added and the mixture was heated and refluxed for 2 days. The THF was removed, the residue was quenched with 10 mL1MHCl, and the crude product was extracted with ethyl acetate (25mL). The ethyl acetate solution was washed twice with 25mL of water, dried by MgSO4, filtered and concentrated. The crude product was purified by column chromatography (eluent cyclohexane/ethyl acetate 2/1v/v). The diethyl ester obtained as described above was hydrolyzed to a dicarboxylic acid ester by reflux heating in 25 mL10 w%NaOH for 4 h. The solution was neutralized with 6MHCl and the solvent was evaporated. Diacid was extracted with 25mL methanol. After the solvent evaporates, 20 mL1MHCl is added and the solution is refluxed for 3 hours. Evaporate the solvent and dissolve the product in 10mL of methanol. Add propylene oxide (5mL) and stir the mixture at room temperature overnight. Filter the precipitate and dry 2-aminoheptanoic acid. |
