In vitro study | PD 128907 HCl is a potent and selective dopamine D3 receptor agonist with an EC50 of 0.64 nM, which is 53 times more selective than that at the dopamine D2 receptor. The use of [PD 128907 in CHOKl-cells is used to study the role of these receptors in the brain, such as inhibiting self-receptors, which in turn restrict dopamine release. |
In vivo study | In normal and γ-butyrolactone (GBL)-treated rats, PD 128907 was effective in reducing DA synthesis. PD 128907 (3 mg/kg) attenuated the toxicity of cocaine overdose and completely inhibited the convulsive and lethal effects induced by cocaine. This protective effect is exerted by a D3 coupled mechanism, which also has a protective effect on seizures. |
