preparation | FMOC-L-aspartic acid prepared as follows: weigh two milligrams of the amino acid derivative into a screw-capped glass vial (1 cm3) and 1 cm3 of 0.1N (about 0.37% w/v) hydrochloric acid in HFIP (10 μl of about 37% HCl in water) was added per 990 μl of HFIP. The solution was allowed to stand and then analyzed by RP-HPLC. The removal of Fmoc-Asp HCl from V-Trt (OtBu)-OH in HFIP/groups was almost instantaneous as judged by the strongly yellow appearance of the trityl cation. After 10 minutes of reaction, no trace of starting material could be distinguished by HPLC, and FMOC-L-aspartic acid was obtained. |
