In vitro study | NS-398 inhibited COX-2 activity in a concentration-dependent manner with an IC50 of 3.8 μm. The NS-398 of 100 μm had no effect on COX-1 activity. NS-398 at 10 μm increased COX-2 and pro-inflammatory cytokine production, induced apoptosis in LNCaP cells, but had no effect in C4-2b cells that did not respond to androgen. C4-2b cells, when treated with NS-398, retain their proliferative and malignant phenotypic characteristics, prompting them to differentiate into neuroendocrine-like cells. Neuroendocrine-like cells produce epithelial and neuronal proteins, and the response of C4-2b cells to NS-398 is regulated by activation of NF-kB transcription factors, NS-398 induced NF-kB signals and down-regulated the expression of ikβ-α protein in LNCaP C4-2b cells. |
