Physical and Chemical Properties | Compared with sildenafil citrate, sildenafil has the advantages of less dosage, very small side effects, and soluble in water and ethanol. Sildenafil mesylate is a selective inhibitor of phosphodiesterase type 5 (PDE5) specific for cyclic guanosine phosphate (cGMP), which can relax the smooth muscle of the penis and quickly erect. |
In vivo study | Long-term administration of sildenafil to young and old rats increased the period of detumescence, and the increase was more significant in old rats than in young rats. In aged rats, the expression and activity baseline of cavernous PDE5 were greater than in young rats. After long-term administration, sildenafil increased the expression of cavernous PDE5 in young mice, but had no effect on old mice. Chronic and acute sildenafil treatment inhibited PDE5 activity in the penis of aged and young mice. |
