In vitro study | In Purkinje fibers, Dronedarone decreased the early and delayed depolarization induced by 1 mM Dofetilide and 0.2 mM Strophantidine. In ventricular myocytes, Dronedarone(10 mM) significantly reduced L-type calcium currents (76.5%) and delayed rectifier potassium currents (97%). [1] In single cells isolated from the sinoatrial node (SAN) of the Rabbit Heart, Dronedarone inhibits I(K(ACh)) by reducing the channel opening probability (from 0.56 to 0.11) channel cell attachment without changing the single channel conductance. [2] In guinea pig ventricular myocytes, when the holding potential is -80 mV, Dronedarone state-dependent inhibition of fast Na(+) channel current, IC50 is 0.7 μm. At, Dronedarone blocked the -40 mV rectangular pulse-induced calcium ion current with an IC50 value of 0.4 μm, while at a holding potential of -80 mV, Dronedarone (10 μm) only 20% of the current is blocked. In vivo study | In the rat normal heart, Dronedarone increases the action potential duration. Dronedarone reduces the late sustained K() current, I(K)(or ISUS) by 69%. Dronedarone induced tonic blockade of I(K). In rats with myocardial infarction, Dronedarone induced a weak increase in fast transient outward currents. [4] |
