| preparation | preparation of 7-methoxy -8-hydroxycoumarin (daphenin -7-methyl ether) (ii-aa): weigh 7,8-dihydroxycoumarin 500mg into a three-mouth bottle, add N,N-dimethylformamide 5mL dissolved sample, add 850mg sodium carbonate, after fully stirring for 15min, methyl iodide is added dropwise by 560 μL, the reaction is carried out at 0 ℃, TLC monitors the reaction process, and the reaction ends after 5h. The reaction droplets are added to 100mL of ice water, hydrochloric acid is added to adjust the pH to be about 3, ethyl acetate is extracted (50mL × 3), ethyl acetate phase is combined, saturated salt is washed twice, and ethyl acetate is removed by decompression distillation of the organic phase, I .e. crude product is separated by silica gel column, the mobile phase is V petroleum ether: V dichloromethane: V acetone = 2:1:0.2. The solvent is removed by vacuum distillation and vacuum drying to obtain 260mg of white solid. |
