preparation | in a 100mL three-neck bottle, add 7.0mL N,N-dimethylformamide, cool to 0~5 ℃, slowly add 1.0mL phosphorus oxychloride dropwise, finish dropwise, continue stirring for 20min, add 1.9g(10mmol) 5-bromoindazole dropwise below 10 ℃ and dissolve in 3.0mL N, the solution of N-dimethylformamide is dropped and reacted at 35 ℃ for 1h. Add 10mL of water under the cooling of ice water, adjust the pH value to neutral with 15% hydroxide, then reflux 20min, cool to obtain yellow solid, and purify the target compound 1H-indazole -5-formaldehyde by silica gel column chromatography. |
