preparation | nitrogen protection, at -78°C, 20ml of 2,3, 5-trichloropyridine (2g,11mmol) solution of tetrahydrofuran was added to lithium diisopropylamide (7.3mL,1.5M cyclohexane, 11mmol) in 10mL of anhydrous THF solution and stirred for another 30 minutes, methyl formate (1.4mL,1.3g,22mmol) dissolved in 14mL THF was then slowly added to the brown solution within 15 minutes, allowing it to slowly heat up to room temperature and stir overnight. The resulting dark brown solution was poured on ice and saturated NaHCO3, extracted with ethyl acetate, washed with brine, dried (Na2SO4) and concentrated. The brown oil was purified by silica gel rapid chromatography and eluted with 20-33% ethyl acetate/hexane to obtain 2,3, 5-trichloro-4-pyridine formaldehyde in yield:(1.7g,74%). |
