Application | 3-nitrophthalic hydrazide can be used to prepare luminol reagents. 3-Nitrophthalic hydrazide has become the most widely used chemiluminescent reagent due to its high luminescence quantum yield and good water solubility, and can react with a variety of oxidants. Its luminescence mechanism is oxidation reaction to emit light. They are catalyzed by horseradish peroxidase under alkaline conditions and oxidized by hydrogen peroxide to produce their excited state intermediates returning to amino benzodiacic acid. When it returns to the ground state, photons will be emitted. Therefore, luminol reagents have good application value and broad market demand prospects. |
preparation | add 100g of dimethyl 3-nitrophthalate, 140g of ethanol and 83.6g of hydrazine hydrate (50%) to a dried 1000ml three-mouth flask, stir and heat to reflux, maintain reflux reaction for 3h, cool down under stirring, cool to room temperature and filter. Crystallization is dissolved and filtered with water, and the solution is acidified with concentrated hydrochloric acid to PH1.5-2, filtered, washed with water and dried to obtain 60g.
