Application | 2,5-dibromopyrazine is a common organic synthon, which can be used as a molecular skeleton to participate in the synthesis of biologically active molecules. |
Preparation | 5-bromo-2-pyrazinamine (D48, step 1)(317 mg, 1.82 mmol) was added to a solution of cooled (ice acetone bath) hydrobromic acid (48% aqueous solution)(2 ml). After stirring for 5 minutes, bromine (0.28 ml, 5.46 mmol) was added, and then an aqueous solution of sodium nitrite (314 mg, 4.55 mmol) was added dropwise within 15 minutes. The mixture is stirred for 30 minutes and heated to room temperature within 30 minutes. Add the solution of sodium hydroxide (2.6g) in water (7 ml) and stir the mixture for 1 hour. The mixture was extracted with dichloromethane. The dichloromethane layer is combined and evaporated in vacuum to obtain the target compound. |
