2-dimethylamino-4-hydroxy-5, 6-dimethylpyrimidine is prepared by the reaction of N,N-dimethylguanidine sulfate and ethyl α-methyl acetoacetate.
Add 4.6g of acid binding agent, alcohol and 14gN,N-dimethylguanidine sulfate into a 250mL three-mouth bottle equipped with a stirrer and a water separator, stir for 2 hours, add 21.2g of α-methyl acetoacetate and solvent, slowly raise the temperature under stirring until the reaction temperature reaches about 140 ℃, keep the temperature for 2 hours, recover the solvent under reduced pressure, dissolve the solid with chloroform and adjust the pH = 7, the content is 92.69% and the yield is 93.43%.
