preparation | A preparation method of 2-acetylfuran: 1) extraction of volatile oil: Take dry aerial parts of perilla, after crushing, the volatile oil was extracted by steam distillation for 6h and dried with anhydrous sodium sulfate to obtain the volatile oil. The volatile oil was stored in the fresh-keeping room of refrigerator at 4 ℃ for standby use. The yield of volatile oil was 0.24%. 2) compound separation: 10ml of volatile oil from Perilla frutescens was purified by normal phase silica gel (160-200 mesh) column chromatography. The specification of silica gel column was 50mm × 500mm, with 5 gradient elution, and 5 flow segments were obtained for each gradient, each stream was segmented into 200ml and the solvent was recovered at 35 °c under reduced pressure. The five gradients were n-hexane, n-hexane-ethyl acetate 80:1, n-hexane-ethyl acetate 50:1, n-hexane-ethyl acetate 20:1, and ethyl acetate. 25 fraction sections were obtained, and the fraction section 9 was separated by normal phase silica gel column chromatography to obtain 2.5g of the compound 2-acetylfuran. |
