introduction
5-bromo-2-methylpyridine is an important pharmaceutical intermediate, which has broad prospects in the production of cardiovascular and cerebrovascular drugs and respiratory drugs, and has huge market potential.
use
2-amino -6-methylpyridine is an important organic intermediate. It is an important raw material for the synthesis of nerve growth factor receptor TrkA inhibitor 2-amino -5-(thioaryl) thiazole. It shows anti-proliferation effect on tumors. It is a precursor for the synthesis of erythromycin derivatives. It has a good effect on Staphylococcus aureus, Streptococcus pneumoniae and Streptococcus pyogenes.
chemical properties
white or yellowish crystals. Melting point 41 ℃, boiling point 208-209 ℃, relative density 1.0432(45/4 ℃). Easily soluble in water, soluble in alcohol, ether, acetone, benzene and other organic solvents. It's hygroscopic.
use
1. Pharmaceutical intermediates. It is used in the production of nalidixic acid, but also in organic synthesis, the manufacture of dyes and photographic developers.
2. Used in organic synthesis, used as an intermediate in medicine and dyes
3, 2-amino-6-methylpyridine is used to make photographic developers, dyes and drugs.
production method
It is obtained by 2-methylpyridinylation. Put the liquid ammonia into the reaction tank, add high-iron nitrate, slowly add small pieces of metal sodium after stirring, and continue stirring for 3 hours after adding. Ammonia is discharged naturally until the reaction liquid is in the form of a slurry to obtain ammonia sodium liquid. Add anhydrous xylene to sodium ammonia, stir and heat up, and evaporate to remove residual ammonia. When the temperature is raised to 120 ℃, 2-methylpyridine is added dropwise, and the temperature is kept at 124-129 ℃ for 10h. Slowly add appropriate amount of water to hydrolyze at 90-100 ℃, stir for half an hour after hydrolysis, stand and separate the xylene layer, distill and recover xylene, and then distill under reduced pressure to collect 90-120 ℃(1.33-2.67kPa) fraction, I .e. 2-amino -6-methylpyridine. 72% yield.
