Preparation | 3, 5-difluoropyridinonitrile (23.19mmol) is dissolved in ethanol under stirring, and then concentrated hydrochloric acid aqueous solution (2.6mL) is added. Palladium (10% on carbon) is added under nitrogen, and then the mixture is hydrogenated at 30psi for 2 hours using the Parr apparatus. The catalyst is removed by filtration and the filtrate is concentrated under reduced pressure. The residue was divided between EtOAc(40mL) and water (40mL). The water layer was separated, then the pH value was adjusted to 9 with 50% aqueous sodium hydroxide solution (3mL-4mL), and extracted with dichloromethane (3 × 40mL). The combined extract was dried (MgSO4) and concentrated under reduced pressure to obtain light green/brown oil (2.5g,75%). 1H NMR(400MHz,DMSO)8.45(1H,d),7.89-7.84(1H,m),3.82(2H, s),3.33(1H,bs), 1.87(1H,bs). |
