Preparation | 2-bromo-3-aminopyridine can be obtained from 3-aminopyridine as raw material through NBS bromination. Take 47 grams (0.5 mol) 3-aminopyridine dissolved in 750 ml (15 times the amount) CH2Cl2, a 1-liter three-mouth bottle, use dry ice and ethanol to control the temperature -10 ℃ (internal temperature), add 89 grams (0.5 mol ) NBS into it in batches for about 15 minutes, and the raw materials almost disappear after 15 minutes of reaction, add water and separate the liquid, the aqueous phase is extracted with dichloromethane, the organic phase is combined to dry, spin dry, using dichloromethane/petroleum ether = 1:3 as the eluent to get 25 grams of 2-brom-3-aminopyridine. |
