Preparation | Preparation of (4-chloro-2-fluorophenyl) methanol: At 0°C, to 4-chloro-2-fluorobenzoic acid in THF(15ml) Add LiAlH4 (130mg) to the solution of 300mg-fluorobenzoic acid. The suspension was stirred at 0°C for 16 hours. Dilute the reaction mixture with EA and potassium sodium tartrate aqueous solution and stir at room temperature for 1 hour. Separate the layers and use water to further wash the organic phase. The combined organic layer was dried by MgSO4, filtered and evaporated in vacuum. The crude product was purified by CC(Büchi Sepacore,5g column, solvent A:DCM, solvent B: 3N ammonia in MeOH, ladder degree (expressed in B%):0 to 5, flow rate: 6.0ml/min) to provide 224mg colorless oil. LC-MS(A)tR = 0.68min;[M H]: not seen. Preparation of 4-chloro-2-fluorobenzaldehyde: MnO2(480mg) was added to a solution of (4-chloro-2-fluorophenyl) methanol (222mg) in MeCN(20ml). Stir the mixture for 24 hours. The mixture was filtered by diatomite, the organic layer was dried by MgSO4 and evaporated in vacuum. The crude aldehyde is used in the next step without purification. LC-MS(A):tR = 0.76min;[M H]: not seen. |
